Title: Analysis of anthocynanins from fruits of leea guineens and flowers of acanthus pubescenes

Abstract: In order to identify and quantify the anthocyanin in flowers of Acanthus pubescens and fruits of Leea guineensis, the anthocyanin pigments were extracted with methanol 0.5 containing tifluoroacetic acid and the crude extract was purified by solvent extraction and amberlite XAD-7. Further purification and isolation was done by sephadex LH-20 column chromatography. Two anthocyanins were isolated from each plant material and their chemical structures were identified by a combination of both chromatographic (TLC, HPLC) and spectroscopic (UV-Visible, NMR. MS) techniques. The major and minor anthocyanin pigments identified in Acanthus pubescens were cyanidin 3-O- (6 –O-a-rhamnopyranosyl-b-glucopyranoside and cyanidin 3-glucoside respectively and the total anthocyanin content was found to be 610 mg/100g of fresh flowers. Peonidin 3.5-diglucoside was the major anthocyanin in Leea guineensis with cyanidin 3.5-diglucoside as the minor pigment with a total anthocyanin content of 979 mg/100g of fresh fruits.
